Multiplex immunoassays are a cutting-edge technology combining the principles of fluorescence immunoassay (FIA)  with the advantages of microarrays to enable simultaneous detection and quantification of multiple target analytes in a single sample.                                                                                                                                     
In the context of autoimmune and infectious diseases, this technology refers to the simultaneous detection and measurement of multiple analytes such as auto-antibodies and antigens.

About Multiplex immunoassays

Microsphere-based fluorescent assays have been developped with the AtheNA Multi-Lyte® Test analyzer, built by Luminex® Corporation on the Luminex® 100/200™ System using xMAP® Technology.

This system uses polystyrene microspheres labelled internally with different ratios of two different fluorochrome (one red and one infrared) to provide a unique spectral signature. Each fluorochrome can have any of the 10 possible levels of fluorescence intensity, thereby creating a family of 100 spectrally addressed bead sets. The antigens corresponding to autoantibodies are bound to the microspheres. Each of the 100 microbeads that can be differentiated by their fluorescence carries a specific immobilized antigen for a single autoantibody.

At the same time, a green laser excites the external reporter fluorescence to quantify the specific reaction related to each autoantibody.
