Use of Capillary Electrophoresis Migration Position for the Presumptive Identification of Hemoglobin Variants Prevalent in China

In this publication, the migration positions of hemoglobin variants prevalent in China were determined by Capillary Electrophoresis and compared with the positions established by two major reference laboratories in the United States and France.

Capillary Electrophoresis (CE) is increasingly used for hemoglobinopathy screening. The authors present the migration positions of hemoglobin (Hb) variants prevalent in China, as determined by CE. These migration positions showed excellent precision and low inter-individual variability.

A total of 107 Hb variants (44 HBA, 42 HBB, and 21 HBD variants) identified in 943 carriers via DNA sequencing, were analyzed using the CAPILLARYS 3 TERA instrument. The migration positions of 56 of these variants were compared with those established in previous publications from Henri-Mondor University Hospital (Creteil, France) and the Mayo Clinic (Rochester, MN, USA).

The authors conclude that the CE migration position serves as a reliable indicator for identifying Hb variants and can help build a well-characterized library of Hb variants, thereby enabling more accurate presumptive identification.
